      以新疆石河子地区月季花为试材,采用Illumina (MiSeq) 平台进行26S rDNA 高通量测序分 析、α 多样性分析、可培养酵母菌的分离纯化技术(稀释涂布法和孢子弹射法)、26S rDNA D1/D 区测 序鉴定、糖发酵及碳同化等生理生化实验,研究了月季花上附生酵母菌(可培养和不可培养)菌群多样 性, 并对系统进化进行分析,以期为有效开发利用新疆酵母菌资源奠定基础,并为探索花蜜微生物与植 物相互作用提供理论依据。结果表明:经26S rDNA 高通量测序筛选后得到41 337 个优化序列,划分入 3 078 个OTU,月季附生酵母菌群落结构丰富,鉴定为3 个门、10 个纲、13 个目、22 个科、37 个属、 90 个种。担子菌门(Basidiomycota)为优势门、占18.9%,线黑粉菌属(Filobasidium)为优势属、占 12.2%,酵母菌种群的丰度存在差异;可培养的酵母菌共鉴定出3 个酵母菌属4 个已知种群,分别为季也 蒙毕赤酵母(Meyerozyma guilliermondii)、粘性红酵母(Rhodotorula mucilaginosa)、乌兹别克斯坦隐球 酵母(Cryptococcus uzbekistanensis)、浅白隐球酵母(Cryptococcus albidus),优势酵母属为隐球酵母属 (Cryptococcus)。

      Using the Rosa chinensis Jacq. in Shihezi area of Xinjiang as the material, the Illumina (MiSeq) platform was used to carry out 26S rDNA high throughput sequencing analysis and Alpha diversity analysis. The experiment made use of isolation and purification techniques of culturable yeast (dilution coating method and spore ejection method), sequencing and identification of 26S rDNA D1/D2 region, carbohydrate fermentation tests, carbon assimilation tests and related physiological tests. The diversity and phylogenetic analysis of epiphytic yeast (culturable and non culturable) on R. chinensis Jacq. aimed at laying a foundation for the effective utilization of Xinjiang yeast resources. It also provided a theoretical basis for exploring the interaction between nectar microorganisms and plants. The results showed that 41 337 optimized sequences were selected by high throughput sequencing of 26S rDNA and divided into 3 078 OTU (Operational Taxonomic Unit). The epiphytic yeast community structure was diverse. Then the epiphytic microorganism community structure was identified as 3 phylum, 10 classes, 13 orders, 22 families, 37 genera and 90 species. The abundance of yeast population were different. Basidiomycota was dominant phylum, which was accounted for 18.9%, among the yeast community. Filobasidium was dominant genera, which was accounted for 12.2%. 4 species culturable yeasts from 3 genera were recognized, including Meyerozyma guilliermondii, Rhodotorula mucilaginosa, Cryptococcus uzbekistanensis and Cryptococcus albidus. The dominant yeast genera is Cryptococcus.
