      为进一步挖掘泥螺的药用价值,进行了泥螺糖胺聚糖光谱分析与生物学功能研究。分别采用KBr 压片法、邻二氮菲-Fe2+氧化法、邻苯三酚自氧化法、MTT 法、体外试验法进行糖胺聚糖红外光谱分析、糖胺聚糖对羟基自由基的清除作用、对超氧阴离子自由基的清除作用、对HepG2细胞生长的抑制作用及抗凝血活性研究。结果表明,该糖胺聚糖中含有-OH(3400 cm-1)、-COO-(1639 cm-1)、-SO2(1421 cm-1)、C-O-C(糖环,1023 cm-1)等基团,这些均为糖胺聚糖的特征基团;糖胺聚糖具有较强的清除羟基自由基的能力,但作用弱于维生素C,具有很弱的清除超氧阴离子自由基的能力;糖胺聚糖对肝癌细胞系(HepG2细胞)有较强的抑制作用;体外试验证实糖胺聚糖可延长新西兰白兔的TT、PT、APTT,但与肝素相比活性明显低弱。

      In order to excavate the medicinal value of Bullacta exarata, spectral analysis and biological functions of glycosaminoglycan from B.exarata were studied. Infrared spectroscopy of glycosaminoglycan was analyzed using KBr pressing plate, the scavenging effects of glycosaminoglycan on hydroxyl radical and superoxide anion by phenanthroline -Fe2 oxidation and pyrogallol autoxidation methods were studied, inhibition effect of glycosaminoglycan on HepG2 cells growth with MTT assay was detected, anticoagulant activity of glycosaminoglycan was studied in vitro experiment. The results showed that the glycosaminoglycan contained-OH (3400 cm-1), -COO-(1639 cm-1), -SO2 (1421 cm-1), C-O-C (sugar ring, 1023 cm-1), etc groups, these were characteristics groups of glycosaminoglycan; the glycosaminoglycan had strong ability of scavenging hydroxyl radicals, but weaker than that of vitamin C, and its ability of scavenging superoxide anion was very weak; glycosaminoglycan had stronger inhibitory effect on liver cancer cell lines(HepG2 cells); the glycosaminoglycan could extended the TT, PT and APTT of New Zealand white rabbits in vitro, but its activity was obviously lower than that of heparin.
