      以海南粗榧愈伤组织进行悬浮培养,考察了培养基、维生素、蔗糖浓度、pH 值、接种量对海南粗榧细胞 悬浮培养体系建立的影响,并利用HPLC 技术检测海南粗榧悬浮细胞和培养基中生物碱的含量。结果表明院海南粗 榧悬浮细胞系培养的最适条件院MS 为基本液体培养基、添加蔗糖35 g/L+VB1 10 mg/L、接种量6%、初始pH 值5.5耀 7.0曰发酵过程中每5 d调节pH 值至6.0,有利于细胞的生长和产物的合成曰细胞的生长呈“S”型曲线,细胞生长周期 为30 d,产物含量在培养25 d时达到最大曰细胞活力呈先急剧上升后缓慢下降的趋势。
英文摘要:
      Using the callus of Cephalotaxus mannii for suspension cultivation, the factors influencing the establishment of C. mannii cell suspension culture system, including culture medium, Vitamin, sucrose concentration, pH, inoculum amount, were investigated. The alkaloid content in cell suspension culture system of C. mannii was determined by HPLC. The results showed that the optimal medium for the cell suspension culture was MS+35 g/L sucrose+10 mg/L VB1, the inoculum size was 6% and initial pH was 5.5-7.0. It was useful for cell growth and harringtonine formatio to adjust pH to 6.0 every 5 days in the experiment. The cell growth basically accorded with the Scurve in one culture cycle, the cycle was 30 days, the greatest products was obtained at the 25th day. The cell viability sharply increased at first and gradually decreased in later.
