摘要: 为明确萝卜裂叶性状的遗传特点,开发与裂叶性状连锁的分子标记。以萝卜“武青一号”作为母本,“新洲花叶”作为父本杂交得到F1,将F1分别套袋自交和回交,产生F2和BC1,探讨裂叶性状的遗传规律;结果表明,F1植株叶形表现为裂叶,BC1(F1ד武青一号”)和F2中叶全裂与叶浅裂的植株分离比分别符合1:1和3:1,说明萝卜叶缘裂刻的表型符合显性基因遗传模式,萝卜叶缘缺裂受单个遗传位点控制。利用BSA与AFLP技术相结合筛选与萝卜叶缘裂刻基因紧密连锁的分子标记,制备的SCAR标记引物HY-18与萝卜叶缘裂刻基因紧密连锁,其遗传距离为3.32cM。上述标记的获得为萝卜裂叶性状的分子标记辅助育种以及裂叶基因的克隆奠定了理论基础。 |

Abstract: In order to identify the genetic characteristics of radish leaf cracking traits and developing a molecular marker which linked to leaf edge traits. Use radish variety“WUQING1#” and “XINZOUHUAYE” as the parents to make the hybrid F1 ,Then we make selfing and backcross of F1 in order to get F2 and BC1. The results showed that the leaf edge of F1 was completely lobed , The Separation ratio of leaf edge lobed to not lobed varieties in BC1 and F2 are 1:1 and 3:1, Indicating that the phenotypic characteristics of radish leaf edge was controlled by a single genetic locus. Use the BSA and AFLP to screen the molecular markers closely linked to the lobed leaf edge gene of radish, and the SCAR marker primer HY-18 was closely linked with the lobed leaf edge gene of radish, and the genetic distance was 3.32cM. The obtained markers laid a theoretical foundation for molecular marker assisted breeding and the cloning of split leaf genes. |
