      【目的】为提高牡丹愈伤组织增殖倍数和质量,减轻增殖过程中的褐化现象,为牡丹愈伤组织增殖提供途径。【方法】以牡丹胚轴、子叶愈伤组织为材料,对不同添加物组合及浓度、不同预培养时间等方法手段进行了探究。【结果】牡丹胚轴愈伤组织继代增殖最佳培养基组合是PVP 1.0g.L-1+IAA 0.2mg.L-1+TDZ 1.0mg.L-1+GA3 1.0mg.L-1,增殖倍数为5.75,牡丹子叶愈伤组织继代增殖的最佳培养基组合是PVP 2.0g.L-1+IAA 0.5mg.L-1+TDZ 0.5mg.L-1+GA3 1.0mg.L-1,增殖倍数为5.50。采用预培养30d的外植体,可有效提高增殖倍数并减轻褐化。1.0 mg/L的GA3有利于抑制愈伤组织增殖过程中的褐化现象,并促进愈伤组织的增殖。呈淡黄绿色表面附着絮状物且质地松软的愈伤组织在继代增殖过程中不易褐化且中后期体积增殖变化明显。【结论】牡丹愈伤组织增殖过程中加入1.0mg/L的GA3和采用预培养30d的愈伤组织,可以获得较高的增殖倍数并有效抑制褐化,呈淡黄绿色且质地松软的愈伤组织是良好的愈伤组织增殖状态。
英文摘要:
      【Objective】In order to improve the multiple and quality on the proliferation of Paeonia suffruticosa., reduce the browning phenomenon in the process of proliferation and provides a way for peony callus proliferating. 【Method】By taking the callus induced from the hypocotyl and cotyledon of peony as the testing materials to study the effects of different combination, concentration of plant hormones and different time of preincubation on the proliferation of Paeonia suffruticosa.【Result】The optimum combination of Paeonia suffruticosa. to proliferate from hypocotyls explants was PVP 1.0 mg.L-1+IAA 0.2 mg.L-1+TDZ 1.0 mg.L-1+GA3 1.0 mg.L-1, the proliferation multiples was 5.75, and the optimum combination of Paeonia suffruticosa. to proliferate from cotyledon explants was PVP 2.0 mg.L-1+IAA 0.5 mg.L-1+TDZ 0.5 mg.L-1+GA3 1.0 mg.L-1, the proliferation multiples was 5.50. Use explants preincubated for 30 days can increase the proliferation multiples and reduce browning. GA3 at 1.0 mg/L is beneficial to inhibit browning and promote callus proliferation. The callus which surface are yellowish with green and have flocs and soft texture were not easy to brown in the process of proliferation and the volume proliferation changed obviously in the middle and late stages.【Conclusion】Adding 1.0 mg/L of GA3 and using the callus was pre-cultured for 30 days during the peony callus proliferation process, can get higher proliferation multiples and inhibit the browning effectively. The callus which yellowish with green and soft in texture is a good state for callus proliferating.
