      【目的】鉴定来自不同产区分离的桃根癌病病原菌土壤杆菌(Agrobacterium tumefaciens)的致病力。【方法】利用根癌土壤杆菌与T-DNA转移和瘿瘤形成相关的virD2和ipt基因进行分子检测,利用3-酮苷试验等生物学手段对不同地区桃根瘤组织中分离的桃根癌土壤杆菌进行致病性鉴定;并利用指示植物检测不同生物类型菌株的致病能力,最后在不同品种桃植株接种后验证其致病性。【结果】virD2和ipt基因两对特异引物在21株病原菌中均出现203 bp和427 bp的目的片段,表明分离的病原菌均属于根癌土壤杆菌。3-酮苷试验结果表明,有3株菌株由蓝变黄是Ⅰ型,其余18株颜色不变,为生物Ⅱ型。指示植物表明,虽然病原菌都能对植株形成瘤,但生物生物Ⅱ型明显要强于生物菌株致病力要强于Ⅰ型,将生物生物Ⅱ型病原菌接种于不同品种桃树均能长出明显的瘤,也验证了生物Ⅱ型病原菌具有更强致病力。【结论】不同产区分离的桃根癌病病原菌土壤杆菌多为生物Ⅱ型,其致病力很强,几乎能侵染所有的桃品种。
英文摘要:
      【Objective】 In this study, bacterial strains were isolated from tumor tissues collected from several peach production regions. 【Method】 The virulence related to t-DNA transfer and gall formation were identified by virD2 and ipt genes,and the 3-ketoglycosides test were used to screen the pathogenicity of Agrobacterium tumefaciens soil bacillus isolated from peach root nodules in different regions. Indicator plants were used to detect the pathogenicity of strains of different biological types. Finally, the pathogenicity of peach plants of different varieties was verified after inoculation biological means. 【Result】 The PCR results showed that there were all 203 bp and 427 bp target fragments of the specific primers virD2 and ipt genes in 21 strains of pathogenic bacteria, it indicating that the isolated pathogens were all belonging to rhizocarcinoma soil bacillus and only 3 strains belonging to biotype Ⅰ. 【Conclusion】The biotype Ⅱ strains had more strong pathogenicity and could infect almost all peach varieties and peach wild relatives.
