      【目的】溪黄草是一种我国民间习用传统中草药。而作为药材商品的“溪黄草”,其基源植物约有3-4种。现有的鉴别和考证研究认为线纹香茶菜是“溪黄草”的正品之一。但是,在线纹香茶菜种内,种质资源的鉴定评价研究较少,尤其是其次生代谢物的差异研究更是鲜有报道。根据性状测试,广泛栽培的线纹香茶菜多为紫花类型,而我们通过系统鉴评获得了一份可稳定遗传的白花型种质。由此,我们利用代谢组学技术开展了两种质的代谢产物比较研究,可为正品“溪黄草”种质创制、新品种选育提供依据。【方法】本研究采用UPLC-MS/MS分析技术,以白花线纹香茶菜种质为研究对象,广泛栽培的紫花线纹香茶菜种质为对照,分析比较二者之间的差异代谢物,并进行了差异代谢物富集通路分析。【结果】从两份种质检测到的868种代谢物中筛选到125种差异代谢物,其中,有67种在白花种质中显著上调,58种表现下调。两种质的差异代谢物主要包括黄酮类、有机酸、氨基酸及其衍生物、酚酸类、生物碱、游离脂肪酸等,其中黄酮类化合物占比最高,达到46%。进一步的通路富集分析结果显示,125个差异代谢物共注释到47条代谢通路,其中,显著富集的有类黄酮(Flavonoid)生物合成,异黄酮(Isoflavonoid)生物合成,花青素(Anthocyanin)生物合成,黄酮和黄酮醇(Flavone and flavonol)生物合成,嘌呤(Purine)代谢,芪类化合物、二芳基庚烷和姜酚(Stilbenoid, diarylheptanoid and gingerol)生物合成等途径。【结论】本研究通过深度鉴评获得的白花型线纹香茶菜种质,其黄酮类、黄酮醇类化合物显著多于紫花型种质,将为后期线纹香茶菜种质资源的挖掘、创制和新品种选育研究提供良好材料。

      【 Objective 】 Xihuangcao is a traditional Chinese herbal medicine commonly used in Chinese folk culture. As a medicinal commodity, "Xihuangcao" has about 3-4 plant species as its source. The existing identification and research suggest that Isodon lophanthoides is one of the authentic products of "Xihuangcao". However, there are few studies on the identification and evaluation of germplasm resources in the varieties of Isodon lophanthoides, especially the differences in secondary metabolites. According to trait testing, the widely cultivated Isodon lophanthoides plants are mostly of the purple flower type and we have obtained a stable white flower type germplasm through systematic evaluation. Therefore, we utilized metabolomics techniques to conduct comparative studies on the metabolites of two germplasm, which can provide a basis for the creation of authentic Isodon lophanthoides germplasm and the breeding of new varieties. 【 Method 】The white flower Isodon lophanthoides germplasm was used as the research object, while the widely cultivated purple flower Isodon lophanthoides germplasm was used as the control. UPLC-MS/MS analysis technology was used to analyze and compare the differential metabolites between the two germplasm, and differential metabolite enrichment pathway analysis was also conducted.【 Result 】125 differential metabolites were screened from 868 metabolites detected in two germplasm samples, of which 67 were significantly upregulated and 58 were downregulated in the white flower germplasm. The differential metabolites between the two germplasm mainly include flavonoids, organic acids, amino acids and their derivatives, phenolic acids, alkaloids, free fatty acids, etc. Among them, flavonoids account for the highest proportion, reaching 46%. Further pathway enrichment analysis revealed that 125 differential metabolites were annotated into 47 metabolic pathways. Among them, significantly enriched pathways include flavonoid biosynthesis, isoflavone biosynthesis, anthocyanin biosynthesis, flavone and flavanol biosynthesis, purine metabolism, stilbenoid, diarylheptanoid and gingerol biosynthesis, etc.【 Conclusion 】The white flower type Isodon lophanthoides germplasm obtained through in-depth evaluation in this study showed significantly more flavonoids and flavonol compounds than the purple flower type germplasm. This will provide good materials for the exploration, creation, and new variety selection of Isodon lophanthoides germplasm resources in the later stage.
