      【目的】水稻稻瘟病抗性基因Pi2和Pita是两个对华南稻区稻瘟病生理小种具有广谱抗性的基因,对于水稻的稻瘟病抗性育种具有非常重要的应用价值,因此开发一套高效的鉴定方法有利于遗传育种和品种改良。【方法】本研究根据高抗稻瘟病品种黄广油占与高感稻瘟病品种广陆矮4号在Pi2基因的第787位、第788位密码子上的变异GCA GGA/GTG TTA,及高抗稻瘟病国际稻种质资源Tetep与高感稻瘟病地方种质资源丽江新团黑谷在第6640位碱基上的变异G/T,基于竞争性等位基因PCR(Kompetitive Allele Specific PCR,KASP)标记技术原理,【结果】利用本研究开发的KASP功能分子标记,可在早期(种子或苗期)检测育种材料的稻瘟病抗性基因Pi2和Pita的等位基因型,预测育种材料的稻瘟病抗性,对育种材料进行准确筛选,不需要将育种材料种到病圃鉴定。【结论】两个稻瘟病抗性基因的功能位点KASP标记能够很好的区分不同抗性的亲本品种,也在育种材料中清楚区分不同的等位基因型,筛选鉴定出携带抗性等位基因的单株,用于后续培育优良的品种,表明功能型KASP标记能够促进抗稻瘟病水稻品种的遗传改良,提高品种培育效率。

      【Objective】The rice blast resistance genes Pi2 and Pita are two genes with broad-spectrum resistance to rice blast physiological races in south China, which have very important application value for rice blast resistance breeding in rice, so the development of a set of efficient identification methods is conducive to genetic breeding and variety improvement.【Methods】 This study according to the high resistance to rice blast varieties yellow oil and high sense rice blast varieties wide dwarf 4 in Pi2 gene 787,788 codon variation GCA GGA / GTG TTA, and high resistance international rice germplasm resources Tetep and high sense of Lijiang new black valley in the 6640th base variation G / T, based on the competitive allele PCR (Kompetitive Allele Specific PCR, KASP) labeling technology principle,【Results】 Using the KASP functional molecular markers developed in this study, we can detect the allele genotypes of the rice blast resistance genes Pi2 and Pita in the breeding materials in the early stage (seed or seedling stage), predict the rice blast resistance in the breeding materials, and do not need to identify the breeding materials in the disease garden.【Conclusion】Two functional loci of rice blast resistance gene KASP markers can well distinguish the different resistance of parental varieties, also clearly distinguish different allele genotypes in breeding materials, screening identified single strains carrying resistance alleles, used for subsequent cultivating good varieties, shows that functional KASP markers can promote resistance genetic improvement of rice varieties, improve the efficiency of breeding varieties.
