      使用ISSR和SSR两种分子标记检测小滴玻璃化法超低温保存牛大力(Callerya speciosa)腋芽后再生植株的遗传稳定性。从100条ISSR 引物中筛选出17条适合引物,共扩增出128条DNA条带。从53 条牛大力SSR 引物中选出8条引物进行银染检测。结果显示,ISSR 和SSR 分子标记,对照和超低温保存后再生植物无差异性条带。两种分子标记的结合充分证明了牛大力在超低温保存后其基因组DNA序列均无变异,因此该材料的遗传稳定性没有受到影响。
英文摘要:
      ISSR and SSR were used to detect genetic stability of cryopreservation of Callerya speciosa axillary buds regenerated plants by droplets-vitrification. 17 suitable primers screened from 100 ISSR-primers were used for amplified and got a total of 128 DNA bands. The 8 primers selected from 53 SSR-primer of C.speciosa were conducted to silver stain. The results showed that blank and cryopreservation treatment were no difference between the bands by ISSR and SSR markers. Accordingly, no nucleotide sequence had altered during the cryopreservation of C.speciosa, and no effect of the genetic stability.
