      建立了双波长法测定常用淀粉原料中直链淀粉、支链淀粉和总淀粉含量的方法。按照双波长测定的等吸收点作图法确定2种淀粉的测定波长和参比波长,直链淀粉的测定波长分别为630、480 nm,支链淀粉的测定波长分别为550、735 nm。结果表明,直链淀粉浓度在0~36 μg/mL、支链淀粉浓度在40~100 μg/mL范围内线性关系良好,符合比耳定律,具有较高的精确度和准确度。常用淀粉原料的总淀粉含量在66.51%~83.65%,其中绿豆淀粉和豌豆淀粉的直链淀粉含量比明显高于其他淀粉原料;糯米粉中的支链淀粉含量最高,其直链淀粉含量过低,导致吸光值太小而检测不出。
英文摘要:
      A new method for the determination of amylose and amylopectin in the commonly used raw materials was established based on dual-wavelength spectrophotometry. According to the principle of dual-wavelength spectrophotometry method, the contents of amylose and amylopectin in the common raw materials of starch were determined at the determination wavelength of 630 nm and 550 nm with the reference wavelength of 480 nm and 735 nm, respectively. The content of amylose and amylopectin could be figured out from two regressive equations. Beer law was obeyed in concentration of 0~36 μg/mL for amylose and 40~100 μg/mL for amylopectin, and both the exactitude and accuracy were very good. The total starch contents of the commonly used raw materials of starch ranged from 66.51% to 83.65%. The amylose contents of mung bean starch and pea starch were significantly higher than other starch raw materials. The amylopectin content of glutinous rice flour was the highest, and the amylose content was extremely low for its absorbance value too low to be detected.
