      根据 14-3-3 蛋白的保守性及菜豆基因组信息,从普通菜豆品种东北小油豆中克隆了一个 14-3-3 蛋白基因 PvGF14h。 生物信息学分析表明,该基因编码的蛋白以 琢 螺旋为主导结构,具有典型的 14-3-3 蛋白 结构域,主要分布在细胞质与过氧化体中,具有磷酸化位点;序列比对及进化树分析表明,它与大豆 14-3-3 蛋 白 SGF14h 有较高的同源性(可达 98.1%);进一步利用实时定量 RT-PCR 分析 PvGF14h 响应冷胁迫的表达模 式,结果表明该基因的表达随着冷胁迫处理时间的增长而增加,因此推测 PvGF14h 可能参与了冷胁迫的信号 转导过程。 这些关于菜豆 14-3-3 蛋白基因的基本信息为进一步研究其功能尤其在响应低温胁迫方面奠定了 基础。
英文摘要:
      A 14-3-3 gene, PvGF14h, was cloned from Phaseolus vulgaris L. based on the conservation of 14-3-3 protein and the genome sequence of common bean. Bioinformatics analysis indicated that the protein encoded byPvGF14h was characterized with alpha-helix predominant structure, typical 14-3-3 domain, phosphorylation sites,and localized at cytoplasm and peroxisome. Sequence alignment and phylogenetic analysis showed that PvGF14h hadthe highest homology with SGF14h from soybean, and the identity was up to 98.1%. Furthermore, real-time PCRanalysis was conducted to examine its expression pattern in response to cold stress. The result showed that thetranscriptional level was promoted by cold treatment, suggesting that PvGF14h might be involved in the signalingtransduction during cold stress. The findings provided some preliminary and important information about the 14-3-3in common bean, which would facilitate to develop further study for understanding its functions, especially inresponse to cold stres
