      运用EMBOSS 及Codon W 等在线软件分析了膜荚黄芪鲨烯合酶(squalene synthase,SS)基因的 同义密码子偏好性,并将其与大豆、拟南芥等7 种植物SS 基因及大肠杆菌基因组密码子偏好性进行比较,为 该基因选择合适的宿主及高效表达提供理论依据。结果表明,膜荚黄芪SS 基因偏好以A 或T 碱基结尾的密码 子,与大肠杆菌基因组共有24 个密码子存在差异。为进一步提高膜荚黄芪SS 基因在大肠杆菌中的表达水平, 需对其密码子进行优化。
英文摘要:
      In order to provide theory basis for selecting appropriate expression systems,synonymou codon bias of squalene synthase(SS)gene of Astragalus membranaceus(AmSS)was analyzed with EMBOSS and Codon W programs,and AmSS with other seven plants and E.coli genome was compared. The results showed that AmSS was bias toward the codons with A and T at the third codon position,and there were 22 significantly different codons between AMSS and SS of E.coli genome. Therefore,codons of AmSS are needed to be optimized to improve its expression level in E.coli.
