      【目的】对广东省大豆资源进行遗传多样性分析,筛选有效的SSR 分子标记,以及构建快速鉴定的DNA 分子身份证。【方法】收集了广东省19 个市37 个县市共96 份大豆种质资源,提取基因组DNA 后,利用30 对SSR 引物进行PCR 扩增,通过测序检测获得相关多态性结果进行聚类分析以及DNA 分子身份证构建,记录相关品种性状并转化为可视化信息。【结果】毛细管电泳检测结果表明,30 对SSR 引物在96 份大豆材料之间均具有清晰稳定的多态性片段,共扩增出273 个多态性等位变异片段,平均每对引物扩增出多态性等位变异片段数14.29 个;不同引物揭示的多态性信息含量(PIC)的范围为0.3891~0.9310,平均值为0.6786,30 对引物可用于区分广东大豆资源。通过聚类分析发现,所收集的大豆样品可以分为3 个类群,具有遗传多样性。从PIC 最高者开始进行引物组合,筛选出6 对能将96 份大豆区分开的引物。【结论】基于6 对SSR 引物扩增结果,对多态性片段排序,通过数字与英文结合编码,成功构建96 份大豆资源的DNA 分子身份证,为广东省大豆种质资源鉴定提供了重要依据。

      The study was conducted to analyze the genetic diversity of soybean resources in Guangdong Province, select effective SSR markers and construct DNA molecular ID for rapid identification.【Method】In this study, 30 pairs of SSR primers were used to amplify the genomic DNA of 96 soybean germplasm resources collected from 37 counties (cities)of 19 municipalities in Guangdong Province. The polymorphic results were obtained by sequencing, and could be used for cluster analysis and DNA molecular ID construction. Related variety traits were recorded and transformed to visual information.【Result】The results of capillary electrophoresis showed that 30 pairs of SSR primers had clear and stable polymorphic fragments among 96 soybean accessions. A total of 273 polymorphic alleles were amplified, and the average number of alleles per primer was 14.29. The range of polymorphism information content(PIC)revealed by different primers ranged from 0.3891 to 0.9310, the average polymorphism was 0.6786. Thirty pairs of primers could be used to distinguish soybean resources in Guangdong Province. Cluster analysis showed that the collected soybean samples could be divided into three groups with genetic diversity. Six pairs of primer combinations were selected according to the value of PIC to distinguish 96 soybean accessions from each other. 【Conclusion】Based on the amplification results of 6 pairs of SSR primers, the polymorphic fragments were sequenced and a set of molecular ID of 96 materials were successfully constructed through combined coding with numbers and English letters, providing an important basis for the identification of soybean resources in Guangdong Province.
