      【目的】提取假芝子实体多糖,研究其粗多糖组成及其抗氧化活性等,为假芝资源开发利用提供参考。【方法】以人工驯化栽培的假芝子实体为材料,采用热水浸提法,经脱蛋白、冷冻干燥获得粗多糖,采用 HPLC、GPC、凯氏定氮法等分析多糖组分、分子量和蛋白质、氨基酸组成,检测假芝多糖清除羟基自由基、ABTS 自由基抗氧化活性,用 MTS 法检测假芝多糖对巨噬细胞 RAW264.7 增殖的影响。【结果】假芝子实体粗多糖为棕褐色粉末,当浓度为 1.5 mg/mL 时羟基自由基、ABTS 自由基清除率分别达 90.26%、83.95%,一定范围内能促进 RAW264.7 巨噬细胞的增殖。假芝子实体多糖蛋白质含量为 16.99%,含有 17 种氨基酸,其中必需氨基酸占 28%,以天冬氨酸和谷氨酸含量最高,二者占比超过 30%。多糖组分由半乳糖、岩澡糖等 10 种单糖组成,其中半乳糖、葡萄糖、甘露糖含量最高,3 种单糖含量占 67.87%。假芝粗多糖数均分子量(Mn)为 28.239 ku,重均分子量(Mw)为 57.502 ku,分布系数 α(Mw/Mn)为 2.036。【结论】假芝粗多糖为含有蛋白质的杂多糖,对羟基自由基、ABTS 自由基清除效果好,为更好的推广应用和科学开发假芝提供了科技支撑。

      【Objective】The polysaccharides were extracted from the fruit body of Amauroderma rugosum, and the crude polysaccharides composition and anti-oxidant activity were analyzed, with a view to providing references for the development and utilization of Amauroderma rugosum resources.【Method】The crude polysaccharides from cultivated fruit body of A. rugosum were obtained by hot water extraction, deproteinization and freeze drying. HPLC, GPC and Kjeldahl method were used to analyze the components, molecular weight, protein and amino acid composition of polysaccharides, and the scavenging ability to hydroxyl radical in vitro, ABTS·+ free radical anti-oxidant activity were explored, the effect of polysaccharides on proliferation of macrophage Raw264.7 was also detected by MTS.【Result】The crude polysaccharides of A. rugosum was brown powder, and its scavenging ability to hydroxyl radicals and ABTS·+ radical were 90.26% and 83.95% respectively at the concentration of 1.5 mg/mL. It could also promote the proliferation of macrophages Raw264.7 in a certain range. The polysaccharide from the fruit body of A. rugosum was a protein-containing(16.99%)heteropolysaccharide, with 17 kinds of amino acid, of which 28% was essential amino acid, and the contents of Asp and Glu were the highest, accounting for more than 30%. The polysaccharides were composed of 10 monosaccharides, including galactose, glucose and mannose. The content of these three monosaccharides was 67.87%. The number-average molecular weight of the crude polysaccharide was 28.239 ku(Mn), the weight-average molecular weight was 57.502 ku(Mw)and the distribution coefficient(Mw/Mn)was 2.036.【Conclusion】The crude polysaccharide from fruit body of A. rugosu is a heteropolysaccharide containing protein, which has good scavenging effect on hydroxyl radical and ABTS·+ free radical. The results of this study provide technical support for the promotion and scientific development of A. rugosum.
