      为了解外源人绒毛膜促性腺激素(human chorionic gonadotrophin, HCG)对黑边石斑鱼(Epinephelus fasciatus)排卵的影响,以成熟的黑边石斑鱼为研究对象,测量其体长、体重和肥满度后,在2018年7~8月,使用HCG激素(剂量为200 IU/kg或500 IU/kg)分别对6尾雌鱼进行成熟诱导。结果表明,在200 IU/kg注射组中,注射HCG 24 h前在5个体中卵母细胞检测为第三次卵黄球前期阶段;注射48 h 后2个体中出现核移动卵母细胞,1个体为第三次卵黄球后期阶段;注射60 h后1个体出现排卵现象,其余5个体均为第三次卵黄球后期阶段。在200 IU/kg的排卵个体中,注射时其卵母细胞的直径为477.0 μm,注射后60 h卵母细胞的直径增加至624.4 μm。此外,经催产、排卵和受精后共获得总卵数16906粒,受精率为68.7%,孵化率为43.0%。但注射HCG剂量为500 IU/kg的黑边石斑鱼排卵失败。研究表明,使用剂量为200 IU/kg的外源HCG激素可能诱导黑边石斑鱼成熟且排卵,但为了提高排卵率、受精率和孵化率,其注射剂量和效应时间还需要进一步调节。

      To understand the effects of exogenous human chorionic gonadotrophin (HCG) hormone on ovulation of the blacktip grouper (Epinephelus fasciatus), the mature fish were used in this study. The total length, body weight, and condition factor were measured and calculated. And then, 6 fish were injected with HCG hormone from July to August in 2018, and the HCG dosages were 200 IU/kg or 500 IU/kg, respectively. In the group of 200 IU/kg, before 24 h of HCG injection, the tertiary yolk stageⅠ(Ty1) was detected in 5 individuals. After 48 h, the migratory nucleolus stage was found in 2 individuals, and the tertiary yolk stageⅡ(Ty2) was detected in 1 individual. After 60 h, the ovulation phenomenon was observed in 1 individual, and the Ty2 was found in other 5 individuals. In the ovulated individual of 200 IU/kg HCG group, the oocyte diameter was 477.0 μm during the injection while it increased to 624.4 μm after 60 h of injection. After the steps of injection, ovulation, fertilization, and hatching, the total numbers of eggs obtained was 16906, and the fertilization rate and hatching rate was 68.7% and 43.0%, respectively. However, the ovulation phenomenon was not observed in the group of 500 IU/kg. In conclusion, the 200 IU/kg usage of exogenous HCG hormone may induce the maturation and ovulation in the blacktip grouper. However, to improve the fertilization rate and hatching rate, the dosage and effective time of injection should be further regulated.
