【目的】虫生真菌在传代过程中易发生菌种退化和活性降低等现象,选择适宜的菌种保藏方法是减少实验室菌种传代的重要措施。【方法】比较了PDA斜面、甘油和砂土法三种不同保藏方法对13种18株虫生真菌的保藏效果。【结果】在3个月的短期保藏中,除砂土保藏的个别菌株外,三种方法都展现出良好的保藏效果,尤其是斜面保藏法表现更佳,其中有15个菌株的菌丝生长速率、菌丝脱氢酶活性和12个菌株的产孢量高于其它保藏方法,菌丝长势良好,色泽正常。在12个月的长期保藏中,甘油保藏法的保藏效果最好,其菌落色泽、菌丝生长速率及产孢量等指标均与保藏3个月时变化不大。在保藏期为12个月的砂土保藏法中,罗伯茨绿僵菌Metarhizium robertsii MrGX0603、球孢白僵菌Beauveria bassiana BbGX7303、淡紫紫孢菌Purpureocillium lilacinum PiGX0201、环链虫草Cordyceps cateniannulata CcGX32S01以及玫烟色虫草Cordyceps fumosorosea CfGX4206表现出较好的保藏效果,其菌丝脱氢酶活性较高,产孢量也较好,分别为53.50×106孢子/cm2、52.23×106孢子/cm2、62.00×106孢子/cm2、54.21×106孢子/cm2、53.79×106孢子/cm2,且菌株的菌丝生长速率、脱氢酶活性、产孢量下降的幅度较小;而18株菌种经斜面保藏12个月后活力指标下降明显,砂土法的保藏效果比斜面法较好。【结论】在相同的保藏方法与时间下,不同菌种的保藏效果表现出明显差异。根据保藏期限的不同可选择不同的保藏方法,PDA斜面保藏法最适用于3个月左右的短期保藏;甘油保藏法在保藏期为1年时保藏效果最好;而砂土保藏法更适合保藏产孢量大于50×106孢子/cm2的菌株。因此,更精细的个性化的菌种保藏方法需要在实践中不断改进。 |

【Objective】 Entomogenic fungi are susceptible to strain degradation and reduced activity during the passage process.. Selecting suitable strain preservation methods is a critical measure to reduce the passage of fungal culture in laboratory. 【Method】 In this study, the preservation efficacy of three different methods—PDA slants, glycerin frozen, and sand tubes—was compared for 13 species and 18 strains of entomogenous fungi. 【Result】 During the short-term preservation period of 3 months, all three methods demonstrated satisfactory preservation effects, with the PDA slant method showing particularly favorable results. Among these, 15 strains exhibited higher mycelial growth rates and dehydrogenase activities, while 12 strains showed higher spore production compared to other methods, with good mycelial growth and normal coloration. Over a 12-month period, the glycerol method yielded the best preservation effect, with minimal changes in colony color, mycelial growth rate, and spore production compared to the 3-month period. In the sand tube preservation method which has a storage period 12 months, Metarhizium robertsii MrGX0603, Beauveria bassiana BbGX7303, Purpureocillium lilacinum PiGX0201, Cordyceps cateniannulata CcGX32S01 and Cordyceps fumosorosea CfGX4206 demonstrated superior preservation outcomes. These strains showed higher dehydrogenase activity in the mycelium and better conidial production. At the 12 months mark, the conidial production was 53.50×106 spores/cm2 for MrGX0603, 52.23×106 spores/cm2 for BbGX7303, 62.00×106 spores/cm2 for PiGX0201, 54.21×106 spores/cm2 for CcGX32S01, 53.79×106 spores/cm2 for CfGX4206, and the mycelial growth rate, dehydrogenase activity and sporulation of the strain decreased slightly. The latter’s vitality of 18 strains had apparently decreased after 12 months of storage, and the effect of sand tubes preservation method was better than that of PDA slants. 【Conclusion】 Under identical preservation methods and timeframes, the preservation effects varied markedly among different strains. Different preservation methods can be selected according to the different storage period. The PDA slant preservation method is most suitable for short-term preservation of approximately 3 months. The glycerol method provided the best preservation effect for a 1-year storage period. The sand tube preservation method is more appropriate for strains with sporulation quantities exceeding 50×106 spores /cm2.Therefore, continuous refinement and personalization of strain preservation methods are necessary in practice. |
