      摘 要:【目的】明确番茄潜叶蛾(Tuta absoluta)嗅觉系统中的气味受体(Odorant Receptor,OR)家族基因的数量及其进化关系;并探讨OR家族基因在番茄潜叶蛾不同发育阶段的表达情况,为新型绿色防治策略提供理论依据。【方法】 本研究基于番茄潜叶蛾的全基因组序列,运用TBtools和BITACORA软件工具,并结合blast同源比对和基于结构域的HMMER分析,对番茄潜叶蛾基因组水平的OR家族基因进行鉴定。进一步采用MAFFT软件对番茄潜叶蛾和其它六种昆虫OR家族基因的代表序列进行比对,并采用IQ-TREE软件开展系统发育分析和利用Image GP分析了番茄潜叶蛾在不同发育阶段各OR基因的表达情况,对相关基因进行qPCR验证。【结果】经过同源基因分析、结构域识别,并过滤不完整的序列;在番茄潜叶蛾基因组中共鉴定出51个OR基因,这些基因的编码序列(CDS)平均长度为1029bp,外显子(exon)平均含有7个片段。在番茄潜叶蛾与其它六种昆虫的系统发育分析中,TabsOR01、TabsOR28、TabsOR34和TabsOR38四个基因聚集于一个明确的分支,而其余基因则散布在其他分支中。在染色体定位方面,这51个TabsOR基因分布于19条不同的染色体上,其中第7号染色体含有最多的OR基因(7个)。对番茄潜叶蛾OR基因的转录组数据在不同发育阶段的表达分析显示,OR基因在成虫期表达最多;其次是幼虫期,在卵期高丰度表达的基因较少各基因在不同发育阶段的表达水平存在显著差异。【 结论】 成功鉴定出51个番茄潜叶蛾OR家族基因,其中有4个基因聚集于一个分支,这些OR基因在番茄潜叶蛾不同发育阶段中的表达模式各异。

      Abstract: 【Objective】To identify the number and evolutionary relationships of odorant receptor (OR) family genes in the olfactory system of Tuta absoluta, analyze their expression patterns across different developmental stages, and provide insights for pest control strategies.【Method】In this study, OR family genes were identified at the genomic level using TBtools and BITACORA software, leveraging the genome sequence of Tuta absoluta. This was combined with BLAST homology searches and structural domain-based HMMER analyses. The representative sequences of the OR family genes of the Tuta absoluta and six other insect species were further compared using MAFFT software, and the phylogenetic analyses were carried out using IQ-TREE software and the expressions of the OR genes of the Tuta absoluta were analysed at different developmental stages by using Image GP, and the related genes were verified by qPCR.【Result】A total of 51 OR genes were identified in the Tuta absoluta genome after homologous gene analysis, structural domain verification, and filtering of incomplete sequences. The coding sequences (CDS) of these genes had an average length of 1,029 bp, with an average of seven exons per gene. Phylogenetic analysis revealed that four genes (TabsOR01, TabsOR28, TabsOR34, and TabsOR38) clustered in a well-defined branch, while the remaining genes were distributed across other branches. Chromosomal localization analysis showed that these 51 TabsOR genes were distributed across 19 chromosomes, with chromosome 7 harboring the highest number of OR genes (7). Analysis of transcriptome data on the expression of Tuta absoluta OR genes at different developmental stages showed that OR genes were most highly expressed at the adult stage; followed by the larval stage, with fewer genes expressed in high abundance at the egg stage.【Conclusion】Fifty-one tomato leaf miner OR family genes were successfully identified, among which four genes were clustered in a single branch, and the expression patterns of these OR genes varied in different developmental stages of Tuta absoluta.
