      【目的】近年来益生菌在水产养殖中的应用越来越广泛,然而卵形鲳鲹源的益生菌还鲜有报道。本研究从卵形鲳鲹肠道中分离到一株粪肠球菌并对其系列特性开展了分析,为该菌株在卵形鲳鲹中的应用提供理论基础。【方法】无菌采集健康卵形鲳鲹肠道内容物,并进行细菌的分离、筛选、鉴定,对典型菌落做形态观察、革兰染色、生化鉴定、16S rRNA基因序列分析、药敏试验、耐盐能力试验、产酸能力以及生长曲线测定和毒力基因鉴定。【结果】根据分离菌株的形态特征、革兰染色结果、生化特性,结合16S rRNA基因序列分析测定结果分析,确定其为粪肠球菌。药敏试验结果表明分离菌株对哌拉西林、多粘菌素B、青霉素、复方新诺明、苯唑西林等药物呈现出不同程度的耐药性。耐盐能力试验结果表明分离菌株可以在NaCl浓度0-8%生长,具有一定耐盐性。产酸能力和生长曲线测定结果表明,随着菌株的大量繁殖,菌液的pH值也快速下降,其产酸能力增强。毒力基因的PCR扩增结果表明,分离菌株包含efaA、agg、as、fsrA这4种毒力基因。【结论】本研究成功从卵形鲳鲹肠道内分离到潜在益生菌菌株,发现其具有良好的耐盐和产酸能力,并且检测到有助于其粘附在肠道上皮细胞的基因,为该菌株的后续应用奠定了基础。

      [Objective] In recent years, probiotics have been increasingly used in aquaculture, but probiotics derived from Trachinotus ovatus are rarely reported. In this study, a strain of Enterococcus faecalis was isolated from the intestine of Trachinotus ovatus and its characteristics were analyzed, which provided a theoretical basis for the application in Trachinotus ovatus culture. [ Method ] Sterile collection of the intestinal contents from the healthy golden pompano(Trachinotus ovatus) was sampled aseptically for bacterial isolation and culture, morphological observation, Gram staining, biochemical identification, 16S rRNA gene sequence analysis, drug sensitivity test, salt tolerance test, acid production ability, growth curve determination, and virulence gene identification of typical colonies. [ Result ] Based on the morphological characteristics, Gram staining results, and biochemical characteristics of the isolated strain, combined with the 16S rRNA gene sequence analysis results , it was identified as Enterococcus faecalis. The results of the drug sensitivity test showed that the isolated strains exhibited varying degrees of resistance to drugs such as piperacillin, polymyxin B, penicillin, compound sulfamethoxazole, and oxacillin. The results of the salt tolerance test showed that the isolated strains can grow at 0-8% NaCl concentrations, and 12% -16% NaCl concentration is not conducive to the growth and reproduction of the isolated strains. The results of acid production capacity and growth curve measurements indicate that as the strain proliferates in large numbers, the pH value of the bacterial solution also rapidly decreases, and the acid production capacity is enhanced. PCR identification of virulence genes showed that the genotypes of the isolated strains include efaA, agg,as, and fsrA. [ Conclusion ] Potential probiotic strains were successfully isolated from the intestine of golden pompano, and the research results can provide a data basis for exploring potential probiotics as alternative antibiotics.
