      研究建立了一种同时检测饲料中泼尼松、甲基泼尼松龙、倍氯米松、氟氢可的松、醋酸可的松等5 种糖皮质激素的液相色谱分析方法,采用乙腈作为提取溶剂,减少了杂质溶出;同时优化色谱条件,减少了背景干扰。选择Agilent Poroshell 120 SBC18色谱柱(3.0 mm×150 mm,2.7 μm),以40%乙腈水为流动相、流速0.60 mL/min、柱温35.0 ℃、紫外检测器(TUV)、波长254 nm 进行检测,结果表明,5种糖皮质激素分离良好,6 min 内即出峰完毕,线性关系良好,相关系数为0.99957~0.99996,回收率为83.80%-94.04%,检测限0.03~0.05 mg/kg。整个试验过程只需提取、离心、吹干、定容,免除了净化步骤,达到了高效、灵敏、低消耗的效果,能满足样品分析要求。
英文摘要:
      An analysis method was established to detect 5 kinds of glucocorticosteroids in feed by UPLC in this article. Acetonitrile was selected as extraction solvent, with less impurities dissolved out. By optimization of UPLC conditions, background interference was reduced. The Agilent Poroshell 120 SB-C18 column (3.0 mm×150 mm, 2.7 μm) was selected. 40% acetonitrile-water was the mobile phase. The flow rate was 0.60 mL/min. The column temperature was 35益. The detector was TUV, and the detection wavelength was 254nm. The results showed that the 5 kinds of glucocorticoids were separated well, outflowing in 6 min. Good linearity was showed, with correlation coefficients of 0.99957~0.99996. The recovery rates were 83.80%~94.04%. The detection limits were 0.03~0.05 mg/kg. The entire trial process including extraction, centrifugal, blow dry and constant volume, without the step of purification, achieved the effect of high performance, flexibility, low consumption.
