为了解fow2基因在尖镰刀菌古巴专化型(Fusarium oxysporum f. sp.Cubense,Foc)侵染香蕉过程中的作用,同时比较Foc的1号和4号生理小种中fow2基因的差异,采用PCR和RT-PCR方法,分别扩增了这2个生理小种的fow2基因,并对测序结果进行生物信息学分析。测序结果显示,以Foc1、Foc4的gDNA为模板,这两个生理小种的PCR扩增产物大小分别为2439、2440 bp;而以cDNA为模板,PCR扩增产物大小分别为1992、1986 bp。分析结果表明,两个基因都含有6个外显子和5个内含子;没有信号肽,也没有跨膜结构;但根据PROSITE蛋白位点分析,2个生理小种的fow2基因存在一些差异。 |

In order to know the role of fow2 gene in the invasion process of banana by the Fusarium oxysporum f. sp.Cubense(Foc),and compare the differences of fow2 gene between the Foc1 and Foc 4, the two fow2 genes were amplified by Polymerase Chain Reaction and Reverse Transcription Polymerase Chain Reaction, then the am-plification products were sequenced, their biological data were analysied. The sequencing result revealed that the full length gene of fow2 genes from Foc1 and Foc4 were 2 439 bp, 2 440 bp and the open reading frames were 1 992 bp, 1 986 bp in length. The two fow2 genes both included 5 introns and 6 exons, had no signal peptide and no transmembrane domain, but the PROSITE results showed that there were several diffrences between them. |
