The process of dissolving Auricularia auricular polysaccharide out effectively according to the
characteristics of its leathery cell wall was studied. This experiment used the combination of high pressure homogenization
and enzyme method, and optimized the extraction process of polysaccharides from A. auricula by using central composite
design method with Minitab statistical software. The results showed that, A. auricular was homogenized under
homogenization pressure 8~10 MPa for 10~12 min as broken pretreatment, then 0.05 mol/L sodium citrate buffer solution
was used as solvent to extract for 2 h at 80. Cooling to 45, 355 U/g cellulase was added and the reaction lasted 1.2 h
in the liquid ratio of 148 in pH 5.0. Then we adjusted pH to neutral with NaOH, and quickly raised the temperature to
85, stayed 1 h for enzyme inactivation. Under above conditions, A. auricula polysaccharides yield reached 15.37%. |
