      以矮牵牛无菌苗叶片作为转化受体材料,建立并优化了农杆菌介导的矮牵牛遗传转化体系。通过农杆菌介导法将水稻反义类黄酮3'-羟化酶基因(F3'H)转入矮牵牛中,抗性植株经PCR 检测均为阳性植株,初步表明反义F3'H 基因已整合到矮牵牛基因组中;对阳性植株进行半定量RT-PCR 检测,与非转基因植株相比,阳性植株中F3'H 基因的表达减弱,表明导入的反义F3'H 基因抑制了内源基因的表达。
英文摘要:
      Agrobacterium-mediated transformation system of Petunia hybrida was estabilished and optimized with P. hybrida leaf as the material. Antisense F3'H gene was transfored into Petunia by Agrobacterium-mediated transformation system. The resistant plants were all positive plants by PCR detection, which indicated antisense F3'H gene had been integrated into Petunia genome. Compared with non transgenic plants, F3'H gene expression in the positive plants detected by semi-quantitative RT-PCR was abate, which showed that the import of antisense F3'H gene inhibited the expression of endogenous genes.
