应用RAPD、AFLP、SSR 等3 种分子标记,对吉富品系尼罗罗非鱼选育群体的多态性进行了分析。在40
条RAPD 引物、19 对SSR 引物、64 对AFLP 引物中,具有多态性、重复性好的引物分别为17 条、19 对和8 对,扩增出
多态性条带数分别为35、92、181 条。结果表明,RAPD、AFLP、SSR 多态性条带比例分别为20.35%、79.64%和58.77%,
平均每个(对)多态性引物可以扩增出多态性片段数目分别为2.06、4.84 和22.63。说明SSR 和AFLP 是研究吉富罗非
鱼选育群体更为有效的分子标记。 |

RAPD, SSR and AFLP assay were applied in polymorphism analysis of Nile tilapia breeding groups. 17
RAPD primers, 19 SSR primer pairs and 8 AFLP primer combinations showed high polymorphism ratio and good
repeatability on Nile tilapia among 40 RAPD primers, 19 SSR primer pairs and 64 AFLP primer combinations, respectively.
The percentages of polymorphic bands generated by RAPD, SSR and AFLP were 20.35% , 79.64% and 58.77% ,
respectively. The average numbers of polymorphic fragments generated by RAPD, SSR and AFLP primers were 2.06, 4.84
and 22.63, respectively. Based on these results, SSR and AFLP were more effective molecular markers for polymorphism
analysis than RAPD in Nile tilapia. |
