      以牦牛的角蛋白关联蛋白 1.1(KAP1.1)基因为研究对象,根据 GenBank 收录的黄牛 KAP1.1 基因的 mRNA 序列(登录号:NM _001105369.1)设计特异性引物,通过反转录 PCR、PCR 以及克隆方法首次获得牦牛的 KAP1.1 完整 CDS 区,将所得序列提交到 NCBI,登录号为 KJ095199,并对其进行生物信息学分析。 结果表明,牦牛 KAP1.1 基因的 CDS 区全长为 501 bp,共编码 166 个氨基酸;编码的蛋白质属于亲水性蛋白质。 二级结构含有 琢 螺旋区、 延伸链、茁 转角和无规卷曲 4 种。 氨基酸序列与黄牛的同源性最高, 具有 High sulphurkeratin-associated protein 家族蛋白的完整结构域。

      Taking Keratin associated protein 1.1 (KAP1.1) gene of yak as the research object, a specific primerwas designed according to the mRNA sequence of cattle KAP1.1 gene (Accession No. NM _001105369.1). Yak scomplete CDS sequences of KAP1.1 were obtained by RT-PCR, PCR and cloning, the sequences were submitted toNCBI (Accession No. KJ095199) and analyzed by bioinformatics. The results showed that, the length of CDSsequences in yak KAP1.1 gene was 501 bp encoding 166 amino acids which belonged to hydrophilic protein. Thesecondary structures were mainly composed of the alpha helix, extended strand, Beta turn and random coil. Theamino acid sequence had the highest homology with cattle, and it had the complete family proteins domain of Highsulphur keratin-associated protein.
