为优化荔枝皮中原花青素的提取工艺,以粤西地区所产的黑叶荔枝果皮为材料,在单因素考察的基
础上,根据Box-Behnken 设计原理,采用Design-Expert 8.0 设计软件,以原花青素得率为指标,进行响应面优化
试验,并通过DPPH 法评价提取物清除自由基的能力。结果表明,荔枝皮的最佳提取工艺为乙醇浓度68%、提取
温度59℃、提取时间93 min、液料比20∶1,提取2 次,在此条件下荔枝皮原花青素的得率为5.26%。优化后原花
青素的得率有所提高,经纯化后提取物清除自由基的能力与Vc 相当甚至更优。 |

LI Huan-qing,DENG Hong,ZHANG Shu,HU Xue-yan,CUI Yue-xin,CHI Xu-ying(Institute of Materia Medica,Guangdong Pharmaceutical University/Guangdong Provincial Key Laboratory of Advanced Drug Delivery,Guangzhou 510006,China)Abstract:To optimize the conditions for extracting procyanidins from pericarp of Litchi chinensis,Heiye growingin western Guangdong was used as experimental material. Based on single factor test and the Box-Behnken designprinciples,using procyanidins yield as the index,a response surface optimization test was established by using Design-Expert8.0 design software. And the efficiency of antiradical of procyanidins extracted from pericarp of Litchi chinensiswas determined by the DPPH assay. The results show that the optimized extraction conditions of procyanidins frompericarp of Litchi chinensis were obtained as follows:ethanol concentration was 68%,extraction temperature was59℃,extraction time was 93 min,solid-liquid ratio was 20∶1 and extracted twice. Under these conditions,the yield ofprocyanidins was 5.26%. The yield of procyanidins increased after optimizing,and the ability of antiradical of the purifiedprocyanidins was similar or even better than Vc. |
