      鱼体内乙酰胆碱酯酶(AChE)活性的降低是水体环境农药污染的重要生物化学标记。以奥利亚罗 非鱼(Oreochromis aurea)肌肉、肝脏和脑组织为材料,研究乙酰胆碱酯酶的组织分布、动力学性质及12 种杀虫 剂(9 种有机磷类、3 种氨基甲酸酯类)对酶的抑制作用。结果表明:脑组织蛋白含量最低(1.53 mg/mL),比活力 最高(1.34 滋mol/min窑mg)。脑组织AChE 的Km值最小(0.160 mmol/L),其次是肌肉AChE(0.227 mmol/L),最高是 肝AChE(0.902 mmol/L)。脑组织AChE 的Vmax最大(2.35 滋mol/min窑mgp),其次是肝组织,最小是肌肉组织(0.63 滋mol/min窑mgp)。12 种农药对肌肉AChE 抑制作用强弱顺序为:敌敌畏>辛硫磷>敌百虫>呋喃丹>乐果>甲基异 柳磷>马拉硫磷>氧化乐果>丁硫克百威>甲胺磷>对硫磷>灭多威,对脑AChE 抑制作用强弱顺序为:敌敌畏>呋 喃丹>灭多威>敌百虫>辛硫磷>氧化乐果>乐果>对硫磷>丁硫克百威>甲基异柳磷>马拉硫磷>甲胺磷,对肝脏 AChE 抑制作用强弱顺序为:呋喃丹>敌敌畏>辛硫磷>甲基异柳磷>灭多威>敌百虫>氧化乐果>丁硫克百威>对 硫磷>马拉硫磷>甲胺磷>乐果。
英文摘要:
      Acetylcholinesterase (AChE, EC3.1.1.7) plays an important role in environmental insecticide detection. Using muscle, liver and brain tissue as materials, tissue distribution and kinetic characters of acetylcholinesterase in Oreochromis aurea and its sensitivity to 12 kinds of insecticides (including 9 organophosphates,3 carbamates) were studied. The results showed that, brain tissue had the highest specific activity (1.34 mol/minmg) while the lowest protein content. Km of AChE in brain tissue was the minimal with 0.160 mmol/L, Km of AChE in muscle was second with 0.227 mmol/L, and Km of AChE in liver was the highest with 0.902 mmol/L. Vmax of AChE in brain tissue was the highest with 2.35 mol/minmgp, followed by liver tissue, and then muscle tissue with 0.63mol/minmgp. Inhibition test showed that 12 insecticides exhibited strong inhibitions on AChE with the order inmuscle tissue as dichlorvos > phoxim > dipterex > furadan > rogor > isofenphosmethyl > malathion > omethoate>carbosulfan > methamidophos > parathion > methomyl, the order in brain tissue as dichlorvos > furadan > methomyl> dipterex > phoxim > omethoate > rogor > parathion > carbosulfan > isofenphosmethyl > malathion > methamidophos, the order in liver tissue as furadan > dichlorvos > phoxim > isofenphosmethyl > methomyl > dipterex> omethoate> carbosulfan > parathion > malathion >methamidophos>rogor
