      几丁质酶广泛存在于自然界的许多物种,并参与几丁质降解、蜕皮、免疫和致病性等诸多功能。采 用同源克隆结合RACE-PCR 的方法,首次从螨类中克隆到二斑叶螨几丁质酶基因(TuChi)的cDNA 全长序列 (GenBank 登录号:JQ041366)。TuChi 全长1 822 bp,包含3' 非翻译区(UTR)为150 bp 和5爷UTR 为52 bp,开放 阅读框(ORF)为1 620 bp,编码539 个氨基酸。预测的相对分子量为60.7 ku,理论等电点为5.99。二斑叶螨几丁 质酶具有几丁质酶典型的结构域,包括1 个N-端信号肽序列、1 个几丁质催化区域、1 个几丁质结合域,以及连 接催化区和结合域之间的连接区域。与其他物种几丁质酶进化树比较分析表明,TuChi 与昆虫几丁质酶家族 Group 1 具有较高的同源性,推测其与二斑叶螨的蜕皮密切相关。

      Chitinases are widely distributed in nature as they are found in many species with different functions including chitin digestion, molting, defense/immunity and pathogenicity. In this study, Tetranychus urticae chitinase gene (TuChi) was first reported from mites using techniques of homological cloning and RACE-PCR. The full length of cDNA sequence was 1 882 bp (GenBank number: JQ041366), containing a 3UTR (untranslated region) of 150 bp, a 5UTR of 52 bp. The ORF (Open Reading Frame) of Tuchi encoding cDNA consisted of 1 620 bp with 539 amino acids residues. Its deduced amino acid composition revealed a calculated molecular weight of 60.7 kua and theoretical PI of 5.99. TuChi protein was composed of N-terminal leading signal peptide, catalytic domain, linker region and C-terminal chitin-binding domain, showing a typical insect chitinase domain structure. Phylogenetic analysis showd that there were high homologies between TuChi and insect chitinase family of Group 1, implicating that TuChi may have an important role during molting stage of T. urticae.
