玫烟色拟青霉IfB01 菌株在25℃、RH85% 下分别在4 种单一培养基(大米、黄豆粉、玉米粉、小
麦粉)和3 种复合培养基(小麦粉∶麦麸1∶1、黄豆粉∶麦麸1∶1、玉米粉∶麦麸1∶1)培养,以产孢量为
指标筛选出较佳的基础培养基配方。通过正交试验验证加入不同浓度的碳源、氮源优化的基础培养基,筛选较
佳的固体培养基配方为:玉米粉培养基+0.2%KNO3+0.1% 葡萄糖+ 水(1∶1.5 = 物料∶水质量比);通过单因
素试验,确定菌株较佳的培养条件是28℃、光照黑暗交替培养14h∶10h、产孢前期湿度为80%、产孢后期湿
度为50%。 |

Paecilomyces fumosoroseus strain IfB01 was respectively cultured in four single medium (rice,
soybean powder, corn powder and wheatmeal) and 3 kinds of composite culture medium(wheat flour∶wheat bran = 1∶
1, soybean flour∶wheat bran = 1∶1 and corn flour∶wheat bran = 1∶1)at 25℃ and RH85% . Using sporulation as the
index,better basal culture medium formula was selected. Through orthogonal experiment,adding different concentrations
of carbon source and nitrogen source to optimize basal medium,better solid culture medium formula was as follows:
corn powder basal medium +0.2% KNO3+0.1% glucose + water(material∶water = 1∶1.5). Through single factor
experiment,the optimum culture conditions of the strain were as follows:at 28℃ and 14h∶10h(l∶d),the humidity
was 80% at early stage of spores production,and the humidity was 50% at late stage of spores production. |
