岗梅是一种具有重要药用价值的植物,在我国主要分布于广西、广东、福建、江西、湖南和台
湾等地。岗梅枝枯病是岗梅生产上的重要病害。根据Koch’s 法则,对岗梅枝枯病病原分离物进行致病性测
定,并利用真菌通用引物ITS1 和ITS4 对该病原菌的rDNA 间隔转录区(ITS)进行扩增和序列分析,结果
显示,在GenBank 上,与该病菌ITS 序列相似度在99% 以上的均是葡萄座腔菌(Botryosphaeria dothidea)。
因此,初步将岗梅枝枯病病原鉴定为葡萄座腔菌(Botryosphaeria dothidea)。 |

Branch blight was a very important disease in Ilex asprella,occurring mainly in Pingyuan,Maoming
of Guangdong province in recent years. Isolation and pathogenicity test of the pathogen of branch blight of I. asprella
were carried out based on Koch’s postulation. For a molecular identification,18S-28S rDNA internal transcribed
spacer(ITS)region was amplified from the pathogen with primers ITS1 and ITS4,obtaining a PCR product of 600
bp. A BLAST search in GenBank revealed the highest similarity(99%-100%)to the sequences of Botryosphaeria
dothidea strains. Therefore,the pathogen causing branch blight of I. asprella was preliminarily identified as
Botryosphaeria dothidea. |
