础上,采用响应面法进行优化设计,得出最佳提取工艺为:提取温度90℃、NaOH 浓度3%、料液比1∶28
g/mL,在此条件下,多糖提取率为0.68%。此外,对多糖抗氧化活性研究结果显示,多糖浓度为10 mg/mL
时,对DPPH 自由基的清除率为68%、其相应的IC50 值为7.62 mg/mL;对·OH 的清除率为45.72%,其相
应IC50 值为12.44 mg/mL,还原力大小为0.263。红外光谱显示该提取物具备一般多糖类物质的光谱特征。 |

To study the optimal alkaline extraction technology of Sepia lycidas muscle polysaccharides,
the extraction temperature,NaOH concentration and ratio of raw material to water were optimized by response
surface methodology design based on single-factor tests. The optimal extraction conditions were extraction
temperature of 90℃,NaOH concentration of 3%,and ratio of raw material to water of 1∶28 g/mL,respectively.
Under these conditions,the extraction yield of polysaccharides was 0.68%. In addition,the antioxidant activity of
polysaccharides was evaluated. When the concentration was 10 mg/mL,the scavenging rate of DPPH was 68% with
the IC50 value of 7.62 mg/mL,and the scavenging ratio of·OH was 45.72% with the IC50 value of 12.44 mg/mL.
The value of reducing capacity was 0.263. Moreover,IR spectroscopic analysis showed the extract had the common
spectroscopic characteristics of polysaccharides. |
