| 假芝是灵芝科假芝属真菌,具有抗氧化、抑肿瘤等作用,提取假芝多糖,研究其子实体粗多糖抗氧化活性及单糖组分等。以人工驯化栽培的假芝子实体为材料,采用热水浸提法,经脱蛋白、冷冻干燥获得粗多糖,采用HPLC、GPC、凯氏定氮法等分析多糖组分、分子量和蛋白质、氨基酸组成,检测假芝多糖清除羟基自由基、ABTS自由基抗氧化活性,用MTS 法检测假芝多糖对巨噬细胞RAW264.7增殖的影响。结果表明,假芝子实体粗多糖为棕褐色粉末,当浓度为1.5mg/ml时羟基自由基、ABTS自由基清除率分别达90.26%和83.95%,一定范围内能够促进RAW264.7 巨噬细胞的增殖。假芝子实体多糖蛋白质含量为16.99%,具有17种氨基酸,其中必需氨基酸占28%,天冬氨酸和谷氨酸含量最高,二者占比超过30%。多糖组分由半乳糖、岩澡糖等10种单糖组成,其中半乳糖、葡萄糖、甘露糖含量最高,这三种单糖含量占67.87%。假芝粗多糖数均分子量(Mn)为28.239 kDa,重均分子量(Mw)为57.502 kDa,分布系数α(Mw/Mn)为2.036。本试验假芝粗多糖为含有蛋白质的杂多糖,对羟基自由基、ABTS自由基清除效果好,本研究结果为更好的推广应用和科学开发假芝提供了科技支撑。 |

| The crude polysaccharides were extracted form the fruiting body of Amauroderma rugosum, and the anti-oxidant activity and monosaccharide composition of the polysaccharides were analyzed. The crude polysaccharides from cultivated fruiting body of Amauroderma rugosum were obtained by hot water extracted, deproteinization and freeze drying. HPLC, GPC and Kjeldahl method were used to analyze the components, molecular weight, protein and amino acid composition of polysaccharides, and the scavenging ability in vitro to hydroxyl radical, ABTS free radical were explored, the proliferation of macrophage Raw264.7 was also detected by MTS. The crude polysaccharides of Amauroderma rugosum is brown powder, it had strong scavenging ability to hydroxyl radicals and ABTS radical which was 90.26% and 83.95% respectively at the concentration of 1.5ml/mg. It also can promote the proliferation of Raw264.7 macrophages in a certain range.The polysaccharide is a protein-containing(16.99%) heteropolysaccharide, it contents 17 kinds of amino acid, of which 28% was Essential amino acid, Asp and Glu twere the highest, accounting for more than 30%..The polysaccharides were composed of 10 monosaccharides, including Galactose, glucose and mannose. The content of these three monosaccharides was 67.87%. The molecular weight of the crude polysaccharide was 28.239kDa ( Mn), 57.502kDa (Mw), and the distribution coefficient (Mw/Mn) was 2.036. |
