      【目的】冬小包脚菇(Volvariella brumalis He sp.nav.)是我国特有的低温草菇品种,探索冬小包脚菇液体菌种发酵培养最优条件,旨在缩减冬小包脚菇菌种生产时间与成本,促进冬小包脚菇标准化、规模化生产。【方法】以冬小包脚菇为研究对象,设置不同碳氮源、温度、酸碱度、接种量水平,对冬小包脚菇液体菌种发酵培养的最优生长条件进行研究,并采用四因素三水平正交试验,得到冬小包脚菇液体菌种发酵培养最佳配方。【结果】冬小包脚菇液体发酵培养最优碳源为葡萄糖,其次为麦芽糖和果糖,菌丝球干质量分别为1.35、1.35、1.33 g/L;最优氮源为发酵豆粕粉,其次为牛肉膏和发酵玉米浆,菌丝球干质量分别为3.76、2.45、1.92 g/L。选取适宜碳源(葡萄糖、果糖)、氮源(发酵豆粕粉、发酵玉米浆)建立 L9 (34) 正交表进行四因素三水平正交试验,得到发酵培养最优配方为:葡萄糖20.0 g,果糖10.0 g,发酵豆粕粉2.5 g,发酵玉米浆2.5 g,KH2PO4 1.0 g,K2HPO4 0.5 g,MgSO4·7H2O 0.5 g,维生素B1 10.0 mg,pH 自然,去离子水1 000 mL。采用该配方进行验证,接种量为3%,于18℃、120r/min条件下培养15 d,菌丝球干质量值达5.79 g/1L。采用优化配方探索其他培养条件,得到最适温度为18 ℃(菌丝球干质量为5.97 g/L)、最适pH为6~7,且当接种量达7%时,发酵效果最佳(菌丝球干质量为6.31 g/L)。【结论】使用上述优化的液体发酵配方培养冬小包脚菇时,大大缩减了冬小包脚菇的制种周期。

      【Objective】The Volvariella brumalis is a unique low-temperature mushroom from China to investigate its liquid optimal fermentation conditions. The aim was to reduce the production time and cost, and promote standardization and large-scale cultivation of Volvariella brumalis.【Method】The optimal growth conditions of liquid fermentation culture of Volvariella brumalis were obtained from the aspects of temperature, pH, inoculum concentration, carbon sources and nitrogen sources. A Four-factors and three-level orthogonal test was conducted using these factors to determine the best fermentation formula.【Result】The result showed that the optimal carbon source for liquid fermentation cultivation of Volvariella brumalis was in the order of glucose, maltose and fructose. The dry mass of the mycelium bulb was 1.35 g/L, 1.35 g/L, and 1.33 g/L, respectively. The optimal nitrogen source was fermented soybean meal powder, followed by beef paste and fermented corn pulp, and the mycelium bulb dry mass was 3.76 g/L, 2.45 g/L and 1.92 g/L, respectively. Suitable carbon sources (glucose, fructose) and nitrogen sources (fermented soybean meal powder, fermented corn pulp) were selected to establish L9 (34) orthogonal table for four factors and three level orthogonal tests to obtain the optimal formula of fermentation and culture. The prescription is glucose 20.0 g, Fructose 10.0 g, Fermented soybean meal powder 2.5 g, Fermented corn syrup 2.5 g, KH2PO4 1 g, K2HPO4 0.5 g, MgSO4·7H2O 0.5 g, vitamin B1 10 mg, pH natural, H2O 1000 mL. The mycelial bulb dry mass reached 5.79 g/L with this formula, and the inoculum was 3% and cultured at 18℃ and 120 r/min for 15 d. Other culture conditions were explored using an optimized formulation, yielding an optimum temperature of 18℃, a mycelium bulb dry mass of 5.97 g/L,and an appropriate pH was from pH 6 to pH 7. And when the inoculum reached 7%, the mycelial bulb dry mass was 6.31 g/L,【Conclusion】The growth time of mycelium in the liquid fermentation formula is shorter than that of solid seed production culture, which greatly reduces the seed production cycle of Volvariella brumalis.
