      【目的】目前大部分研究的菌株都是Colletotrichum gloeosporioides,因此需要更加全面地了解葡萄炭疽病的病原种类及优势种群,明确其致病力分化情况,为葡萄炭疽病的绿色防控提供参考。【方法】利用组织分离法从葡萄果实发病部位分离病原菌,对病原菌的形态特征、分子生物学信息、致病性、生物学特性进行研究。【结果】从葡萄果实发病部位分离获得6株形态特征基本一致的病原菌,并通过代表菌株进行柯氏验证,证明其具有致病性,为该病害的病原菌。结合病原菌的形态学特征及基于ITS、TUB和GAPDH基因序列构建系统发育树,确定病原菌为隐秘刺盘孢菌(Colletotrichum aenigma)。通过对病原菌的生物特性进行分析,结果显示该病原菌的最佳培养温度为28 ℃,并且此条件下产孢量最多;最佳生长 pH为6;最佳培养碳源为麦芽糖;最佳培养氮源为牛肉浸膏。【结论】隐秘刺盘孢菌C. aenigma也是引起葡萄炭疽病的一种重要病原真菌,在对其展开更深层层次的生物功能分析时,可选择成分为麦芽糖、牛肉浸膏、pH=6的培养基于28 ℃环境下进行培养。

      Abstract:【Objective】 Most of the strains studied are Colletotrichum gloeosporioides, so it is necessary to understand the pathogen species and dominant population of grape anthracnose more comprehensively, and clarify its pathogenicity differentiation, so as to provide reference for the green control of grape anthracnose. 【Method】The morphological characteristics, molecular biology information, pathogenicity and biological characteristics of pathogenic fungus were studied by tissue separation method. 【Result】6 strains of pathogenic fungus with similar morphological characteristics were isolated from the pathogenic parts of grape fruits, and the representative strains were verified by Coriolis to prove that they were pathogenic bacteria of this disease. The pathogen was identified as Colletotrichum aenigma based on the morphological characteristics of the pathogen and phylogenetic tree based on ITS, TUB and GAPDH gene sequences. Through the analysis of the biological characteristics of the pathogen, the results showed that the optimal culture temperature of the pathogen was 28 ℃, and the sporulation quantity was the highest under this condition. The optimum growth pH was 6. The best culture carbon source was maltose. The best nitrogen source was beef extract. 【Conclusion】C. aenigma is also an important pathogenic fungus causing grape anthracnose. In the deeper biological function analysis of C. aenigma, the culture of maltose, beef extract and pH=6 can be selected at 28 ℃.
