      尾部神经分泌系统(CNSS)是硬骨鱼类特有的神经分泌系统.其主要分泌物为促肾上腺皮质激素释放因子(CRH)尾紧张素Ⅰ和尾紧张素II(UⅠ、UⅡ)。选取平均体重为600 g的日本牙鲆(Paralichthys olivaceus),系统研究了日本牙鲆尾部神经分泌系统日节律的变化。荧光定量PCR分析表明,昼夜变化过程中,UⅠ和UⅡmRNA表达量没有显著变化;CRH mRNA表达量在6:00~12:00 显著升高袁12:00后降低。余弦法分析表明,CRH、UⅠ和UⅡmRNA表达峰值依次出现在12院:00、18:00、12:00。免疫组织化学染色结果显示,CNSS 分泌细胞要要Dahlgren细胞中CRH,UⅠ和UⅡ抗血清免疫反应阳性细胞总数目均无显著性差异,均无日节律现象。酶联免疫反应及放射免疫反应检测血浆中激素含量表明袁血浆中CRH、UⅠ和UⅡ、促肾上腺皮质激素(ACTH)和皮质醇(Cortisol)含量在光照-黑暗循环中均没有显著性差异,但UⅠ、UⅡ和ACTH 含量随昼夜交替呈先升高后降低的趋势。上述结果表明,日本牙鲆尾部神经分泌系统各基因存在转录振荡,有一定的日节律变化,激素分泌量亦随日周期发生改变。这证明CNSS在牙鲆适应环境变化过程中发挥重要调节作用。

      The caudal neurosecretory system (CNSS) is a unique neuroendocrine structure for environmental adaptation in fish, and is the major site of expression and secretion of corticotropin-releasing hormone (CRH), urotensin I (UⅠ) and II (UⅡ). This study examined daily changes of mRNA expression and the secretion profile in the CNSS of adult Japanese Flounder. Real Time-PCR result showed that under diurnal cycle, the expression of CNSS UⅠand UⅡmRNA was no significant difference; however, the expression of CRH mRNA was significantly higher at 12:00 than that at 6:00, after 12:00 CRH mRNA had a decreasing tendency. The calculated acrophase of CRH, UⅠand UⅡmRNA in the CNSS were recorded at 12:00, 18:00, 12:00, respectively. Moreover, immunocytochemistry revealed that total numbers of CRH, UⅠand UⅡimmunoreactive cells did not show significant daily differences. Plasma hormone was determined in triplicate by enzyme-linked immuneosorbent assay(ELISA) and radioimmunoassay(RIA). There was no statistical difference about plasma CRH, UⅠ, UⅡ, cortisol and adrenocorticotrophic hormone (ACTH) between daytime and nighttime, however, plasma UⅠa, UⅡand ACTH level went up at the beginning and then dropped till the end. These results suggest that the hormone secretion and gene expression in caudal neurosecretory system of Japanese Flounder has daily rhythm. These results reveal the importance of CNSS in the process of Japanese flounder’s adaption to environmental changes.
