      :以干燥的黑胡萝卜粉为原料,在单因素试验的基础上,选取液固比、pH 值和浸提次数为自变量,色素 提取率为响应值,根据响应面Box-Benhnken 试验设计原理,采用三因子三水平的分析法模拟得到二次回归方程 的预测模型,优化黑胡萝卜色素酶法提取工艺。结果表明,建立的回归模型具有高度显著性,方程对试验拟合度 好,适用于黑胡萝卜色素提取率的预测;各因子对提取率的影响程度依次为:浸提次数> pH 值> 液固比;二次项 对黑胡萝卜色素提取的影响都达到了极显著水平;但响应面分析图表明各因素之间的交互作用均不显著;在纤维 素酶浓度0.5%、浸提温度50℃的条件下,纤维素酶辅助提取黑胡萝卜色素的最佳工艺条件为:液固比30∶1,pH 值3.9,浸提次数2 次。 黑胡萝卜色素提取率的实际值为0.9485 mg/g,与模型预测值0.9508 mg/g 没有显著差异。
英文摘要:
      To optimize the cellulase-assisted extraction process of pigment from black carrot,dried powder of blackcarrot was used as material,on the base of single factor test,response surface methodology was used to investigate theeffects of processing parameters,including ratio of liquid to material,pH value,and extraction time on the extraction rateof pigment,and mathematical model was established by implementing three factors and three levels. The regression modelwas highly significant,and the established regression equation for the extraction rate of pigment had excellent goodnessof fit,therefore the pigment from black carrot could be analyzed and predicted by the model. Factors influencing theextraction of pigment from black carrot were in the order as follows:extraction numbers > pH value > ratio of liquid tomaterial. The quadratic term also reached great difference. But the response surface plots showed that the interactionamong each factor was not significant. When the cellulase concentration was 0.5% and temperature was 50℃,theoptimum conditions were as follows:30∶1 of the ratio of liquid to material,3.9 of pH value,2 of extraction times. Theextraction rate of pigment from black carrot was up to 0.9485 mg/g,which was highly consistent with the predictive valueof 0.9508 mg/g.
