An analysis method was established to detect 5 kinds of glucocorticosteroids in feed by UPLC in this article. Acetonitrile was selected as extraction solvent, with less impurities dissolved out. By optimization of UPLC conditions, background interference was reduced. The Agilent Poroshell 120 SB-C18 column (3.0 mm×150 mm, 2.7 μm) was selected. 40% acetonitrile-water was the mobile phase. The flow rate was 0.60 mL/min. The column temperature was 35益. The detector was TUV, and the detection wavelength was 254nm. The results showed that the 5 kinds of glucocorticoids were separated well, outflowing in 6 min. Good linearity was showed, with
correlation coefficients of 0.99957~0.99996. The recovery rates were 83.80%~94.04%. The detection limits were 0.03~0.05 mg/kg. The entire trial process including extraction, centrifugal, blow dry and constant volume, without the step of purification, achieved the effect of high performance, flexibility, low consumption. |
