In order to excavate the medicinal value of Bullacta exarata, spectral analysis and biological functions of glycosaminoglycan from B.exarata were studied. Infrared spectroscopy of glycosaminoglycan was analyzed using KBr pressing plate, the scavenging effects of glycosaminoglycan on hydroxyl radical and superoxide anion by phenanthroline -Fe2 oxidation and pyrogallol autoxidation methods were studied, inhibition effect of glycosaminoglycan on HepG2 cells growth with MTT assay was detected, anticoagulant activity of glycosaminoglycan was studied in vitro experiment. The results showed that the glycosaminoglycan contained-OH (3400 cm-1), -COO-(1639 cm-1), -SO2 (1421 cm-1), C-O-C (sugar ring, 1023 cm-1), etc groups, these were characteristics groups of glycosaminoglycan; the glycosaminoglycan had strong ability of scavenging hydroxyl radicals, but weaker than that of vitamin C, and its ability of scavenging superoxide anion was very weak; glycosaminoglycan had stronger inhibitory effect on liver cancer cell lines(HepG2 cells); the glycosaminoglycan could extended the TT, PT and APTT of New Zealand white rabbits in vitro, but its activity was obviously lower than that of heparin. |
