Different extraction and detection methods of 1-Deoxynojirimycin in silkworm were compared and screened. The extraction
rate of 1-Deoxynojirimycin from silkworm using 0.05 mol/L aqueous hydrochloric acid solution was higher than using 80% ethanol solution and 25% ethanol-hydrochloric solution. The pH of extract had great influence on derivation reaction, and the proper pH of extract was pH>1.6. Detection sensitivity of fluorescence detector for DNJ was higher than UV-detector. Detection of DNJ was performed with 1.0 mL/min flow rate at 25℃ by using a mobile phase of acetonitrile:0.1% aqueous acetic acid (40:60, V/V). Excitation and emission wavelengths of the fluorescence detector were at 254 and 322 nm, respectively. The injection volume was 10 μL. The calibration curve was linear in the range of 2.5~60 μg/mL, R2 was 0.99996. The average recovery was 101.1652%,RSD was 2.7116% (n=9). |
