Abstract:
      Separating regeneration system into dedifferentiation and redifferentiation processes, we investigated the effects of different medium and hormones on callus induction under dark condition and adventitious buds from induced callus by using the leaves and stem segments of Poplar 84K as explants. The results showed that the efficient medium for callus induction from Poplar 84K leaves was MS+KT 0.5 mg/L+2,4-D 0.2 mg/L; for stem segments it was WPM+KT 0.5 mg/L+2,4-D 0.1 mg/L. The efficient medium for callus-induced adventitious buds was WPM+TDZ 0.05 mg/L+6-BA 0.5 mg/L. This study provided an efficient regeneration system, which would be beneficial for the establishment of an efficient transgenic system of Poplar 84K in the future.
