ISSR and SSR were used to detect genetic stability of cryopreservation of Callerya speciosa axillary buds regenerated plants by droplets-vitrification. 17 suitable primers screened from 100 ISSR-primers were used for amplified and got a total of 128 DNA bands. The 8 primers selected from 53 SSR-primer of C.speciosa were conducted to silver stain. The results showed that blank and cryopreservation treatment were no difference between the bands by ISSR and SSR markers. Accordingly, no nucleotide sequence had altered during the cryopreservation of C.speciosa, and no effect of the genetic stability. |
