To study the extraction conditions of polysaccharide from sea grape, total content of crude polysaccharide, as a indicator,was determined by anthrone-sulfuric acid method, and the extraction technology of polysaccharide from sea grape was optimized with L9(33)orthogonal experimental design and accelerated solvent extraction (ASE) process. Results showed that the optimum extraction conditions were 120℃, 2 times with 35 min each time. The extraction temperature was a significant factor. Under the optimal conditions,the yield of polysaccharide from sea grape reached as high as 51.03 (±0.62)%. Compared to traditional extraction technology of water extraction and alcohol precipitation (30.21% extraction yield), the extraction technology in this paper was simple, rapid, energy-saving and efficient, it was a fast, economical, practical and new technology. |
