In order to obtain the best extraction process for enzyme soluble tilapia scale collagen, response surface methodology was used to optimize the extraction process. On the basis of single factor test results and according to central composite design experimental design principles, a mathematical model was established to describe extraction yield as a function of enzyme substrate ratio, time,temperature, and liquid-solid ratio. The results showed that enzyme substrate ratio, time and temperature had significantly effect on the extraction yield, and the optimal extraction process obtained was as follows: enzyme substrate ratio 3.4%, time 4.0h, temperature 59℃, and liquid-solid ratio 35:1. Under these conditions, the extraction yield was 13.12%, which has no significant difference between the experimental and predicted values. The mathematical model obtained will provide a basis for the extraction of enzyme soluble tilapia scale collagen. |
