The extraction of genomic DNA of two wild macrofungi obtained from Wulashan was carried out by CTAB
method. Then, on the basis of PCR, ITS regions of DNA of the macrofungi were amplifiedby using the special pairs of
primers ITS1F/ITS4 and sequenced. The sequences were compared with those registered in GenBank database, results of
which showed that two fungi belonged to the genus Pholiota sp. and Strophariaceae sp.. Genbank accession numbers of the
ITS gene was FJ596817.1 and JQ627593.1. Then the phylogenetic trees were constructed by neighbor-joining method and
genetic distance was analyzed in UNITE Database (s). The results showed that two wild fungi had a high genetic distance,
one with Pholiota tuberculosa (Pers.) Kuyper & Tjall.-Beuk (UDB015324), and the other with Stropharia coronilla (Fr. ex
Bull.) Qul.(UDB015429, UDB015556), which was up to 5.0. On the basis of the sequence analysis, two wild fruiting bodyforming
fungi were identified as Pholiota tuberculosa and Stropharia coronilla |
