In order to research the relationship between gene F25 and Monopterus albus sex reversal, firstly, we used
quantitative PCR technology to analyze the gene expression in different gonadal tissues, then used situ hybridization technique to
synthesize cRNA probes and locate the F25 gene differential expression at different periods. The results showed that the gene
expressed little in ovary at stage II and III, and began to increase rapidly in ovary at stage IV, meanwhile, its expression also
showed different degrees with the ovarian maturation, ovulation, degradation, its expression reached maximum in the testis. By
positioning analysis, we found that the gene was not essential in the early ovary and mainly expressed in somatic cells in ovary at
stage of IV and V, whereas there was no F25 gene expression in germ cells of the testes, which indicated that F25 gene may play
a role in the process of sex reversal and affect on the somatic gonad tissues. |
