Abstract:
      In order to study the cultivation techniques for virus-free seedling of sugarcane, taking sugarcane 03-75 strains as materials, after the water bath of 52(依0.2)益and culturing, the induction culture of sugarcane leaves, callus differentiation, screening of rooting medium, and the methods of acclimatization, transplanting and virus detection for virusfree seedlings were discussed. The results showed that the method of heat treatment combined with seedling leaves detoxication could remove sugarcane Mosaic, perennial root dwarf disease and other diseases effectively. The hormone types and concentrations were different in different stages, the best medium for callus induction and subculture was MS+2,4-D 3.0 mg/L+NAA 0.1 mg/L, for callus differentiation was MS+ BA 1.0 mg/L + NAA 0.1 mg/L, and 1/2MS+NAA 4.0 mg/L + MET 1.0 mg/L was good for high rooting rate and root stout. By using molecular biology techniques (PCR) for sugarcane Mosaic disease detection and enzyme linked immunosorbent (ELISA) for sugarcane perennial root dwarfing detection of tissue culture plants, the results were negative
