Abstract:
To extract the essential oil from Callistemon viminalis leaf by the method of microwave -assisted hydrodistillation, the extraction conditions were optimized through orthogonal test, and antibacterial activity of the essential oil studied, the inhibition zone, minimal inhibitory concentration (MIC) and minimal bactericida concentration (MBC) against 7 species of human pathogenic bacteria were tested by disk diffusion method and microtiter plate double dilution method. The results showed that the optimum conditions of extraction were as follows: ratio of water to leaves 8颐1 (V/W), microwave power 250 W, extracting time 1.0 h, soaking time 1.5 h, with the extraction yield of 0.793%. The results also showed that the essential oil from C. viminalis exhibited broad spectrum effect against Gram-positive (Bacillus subtilis, Staphlococcus aureus and S. epidermidis) and Gram -negative bacteria (Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumonia and Salmonella paratyphoid B) with the range of 9.26~17.35 mm for inhibition zone and the range of 0.03125~0.0625 g/mL for MIC.
