Cloning and sequence analysis of actin genefragment from Eichhornia crassipes |

Abstract: |
Actin gene fragment from Eichhornia crassipes was amplified with degenerate primers designed according
to the conservative sequences of plant actins by RT-PCT method. It was linked to pMD19-T vector and transformed to E.
coli and then sequenced. Sequence analysis showed that the degree of similarity of this sequence and sequences from
Elaeis guineensis and Musa acuminate etc. reached above 85%. Phylogenetic analysis also revealed that the sequence from
E.crassipes was nearer relative to sequences form some monocot plants such as E.guineensis and M.acuminate etc., but it
could not be clustered into one single group with some plant sequences. The sequence of this study can provide an internal
control gene for further study of other gene expression of E.crassipes. |
