Propagation of Cryptocaryon irritans on Paralichthysolivaceus and observation of its life cycle |

Abstract: |
In order to conserve a strain Cryptocaryon irritans isolated from Paralichthys olivaceus in ChangLe of
Fujian province, the parasites were propagated in laboratory by using P. olivaceus as host, controlling water temperature
between 22益and 24益. The morphological characteristics in each life cycle period were observed and recorded. The
hatchability of tomonts and infection ability of theronts were compared between two groups, as tomonts in one group were
stored in 5益for two months before hatching in 27益and directly hatched in 27益in the other group. Serum collected from
the fish infected after 2 weeks and 4 weeks was detected on clumping and immobilization to theronts. Results showed that
P. olivaceus could be a model host for C. irritans. Up to 40 consecutive cycles were successively established, 8-10 d in one
cycle. Some trophont-like round protoplasts were observed orbiting in the tomont. The hatchability of tomonts dropt to 7.6
% in the first group. And none trophont could be detected on the fish after exposed to 1 000 theronts in this group. The
immobilization titers of serum antibody to theronts could be 1颐320 and 1颐1 280 in two-week infected fish group and fourweek
infected fish group. This indicated that humeral immunity played an important role against C. irritans infection. |
