Based on single factor tests, the response surface method (RSM) was used to optimize the determinationcondition of total saponins in Camellia chrysantha by spectrophotometry. The optimal conditions were as follows: the amount of 5% vanillin -glacial acetic acid was 0.35 mL, the amount of perchloric acid was 1.0 mL, the reaaction temperature was 80益, the reaction time was 25 min and wavelength was 540 nm. Under the optimal conditions, the calibration curve was linear in the range of 0 -114.2 滋g, The relative coefficient was 0.9991. The relative standard deviations of precision and stability were 1.36% and 2.08%, respectively. The average recovery rate was 98.22%. |
