In this paper, we studied effects of six kinds of protectants, including glycerol, sucrose, raffinose, dextrose,sodium glutamate, meso-inositol, on shelf-life of Bdellovibrio sp. BDS01, and then picked out three kinds of protectants with good effect to design orthogonal experiment for the best formula. With Arrhenius equation, the shelf-life of BDS01 microbial preparation was predicted when its concentration dropped down by 2 log, and then experiments were conducted to verify the predicting result. The results showed that the effects of 15% glycerol, 5% sucrose and 5% sodium glutamate were better than others. After testing, 17% glycerol, 5% sucrose, 7% sodium glutamate were optimized by orthogonal experiment.Results of constant temperature verification experiment showed that the real shelf-life of BDS01 at 25℃was constant withthe prediction result and the real shelf-life at 10℃and 4℃were both slightly lowed than the prediction results when BDS01 reduced 2 log. And the real shelf-life of BDS01 reducing 2 orders of magnitude at 4℃was about 13 weeks. |
